Application of the Gyrolab® microfluidic platform to measure picomolar affinity of a PD-L1-binding Adnectin™ radioligand for positron emission tomography

This case study demonstrates sub-nanomolar affinity determination anti-PD-L1 Adnectin to its ligand by scientists at Bristol Myers Squibb (BMS) using the Gyrolab® XP with Affinity module multi-curve fitting software. The authors demonstrated advantages of the miniaturized Gyrolab immunoassay platform to perform thorough yet rapid assay development and measurement of in-solution affinity. Using Gyrolab, the authors were able to determine the affinity for the anti-PD-L1 Adnectin as 13 pM, which previously only had been estimated using SPR to be < 35 pM.
